Especially around (i) how to manufacture appropriate mRNA, (ii) how to get it into target cells, (iii) how transcription occurs with the payload mRNA despite whatever the cell happens to be making itself (in the case of defects in the cellular mRNA; I presume where the relevant DNA strand is unread, there’s no relevant mRNA to muddy the waters). As mRNA is a huge molecule (e.g. not druggable in the conventional sense) the vector will be really interesting to understand. How does it pass through the cellular membrane?
